Changes in cerebellar activation pattern during two successive sequences of saccades

The changes in the cerebellar activation pattern of two successive fMRI scanning runs were determined for visually guided to‐and‐fro saccades in 12 healthy volunteers familiar with the study paradigm. Group and single subject‐analyses revealed a constant activation of the paramedian cerebellar vermis (uvula, tonsils, tuber, folium/declive), which reflects constant ocular motor activity in both runs. A significant decrease in activation of the cerebellar hemispheres found in the second run is best explained by either a decrease in attention or the effects of motor optimization and learning. Benzodiazepine- and alcohol-induced ataxias in rodents have been proposed to be affected by the gamma-aminobutyric acid type A (GABAA) receptor alpha 6 subunit, which contributes to receptors specifically expressed in cerebellar granule cells. We have studied an alpha 6 -/- mouse line for motor performance and drug sensitivity. These mice, as a result of a specific genetic lesion, carry a precise impairment at their Golgi-granule cell synapses. Actin dynamics has been recognized as an important determinant of neuronal migration, and we recently found that the actin-binding protein profilin1 is relevant for radial migration of cerebellar granule neurons (CGN). Electrical stimulation of the cerebellar cortex by implanted electrodes has been shown to ameliorate refractory epilepsy. We investigated the potential therapeutic role of high-frequency cerebellar rTMS in patients affected by refractory epilepsy due to single or multiple foci. Six patients, three with single and three with multiple epileptic foci, underwent 20 rTMS sessions. ERPs to pitch changes: a result of reduced responses to standard tones in rabbits.

EVENT-RELATED potentials (ERPs) were recorded in rabbits when pitch deviant tones occurred in a series of standard tones (oddball situation). In control recordings, the deviant tones were presented without the standard tones (deviant-alone situation). In the oddball situation, significant difference ERPs (deviant ERPs - standard ERPs) could be found in the hippocampal and cerebellar recordings but not in the visual cortex. All the ERPs to the deviant stimuli observed in the oddball situation were also present in the deviant-alone situation. The difference ERPs were therefore based on reduced responses to the standards. A control circuit is proposed to model the command of saccadic eye movements. Its wiring is deduced from a mathematical constraint, i.e. the necessity, for motor orders processing, to compute an approximate inverse function of the bio-mechanical function of the moving plant, here the bio-mechanics of the eye. This wiring is comparable to the anatomy of the cerebellar pathways. A predicting element, necessary for inversion and thus for movement accuracy, is modeled by an artificial neural network whose structure, deduced from physical constraints expressing the mechanics of the eye, is similar to the cell connectivity of the cerebellar cortex. Clinical observations and data from animal experiments point to a physiological facilitatory influence of the deep cerebellar structures on the motor system through the cerebello-thalamo-cortical pathways.
